The identification of autoantibodies in patients with lupus erythematosus

Introduction. One of the laboratory criteria for diagnosis of lupus erythematosus is the presence of antibodies against nuclear antigens detected with the indirect immunofluorescence with the substrate combination Hep-2010/liver. Western blot method can be used to verify the results of Hep-2010 cells.
Aim of study. Determination of the type of autoantibodies present in the serum of patients diagnosed with lupus erythematosus using Western blot method.
Material and methods. 16 serums from patients with different forms of lupus erythematosus were subjected to analysis: SLE (9 cases), DDLE (1), SCLE (4), DLE (2). The preliminary identification of autoantibodies as well as their titer was determined on Hep-2010 cells using IIF method. Western blot test has been used to verify the results of IIF method.
Results. In patients clinically diagnosed with SLE, the following autoantibodies were identified using Western blot: RNP/Sm – 3 patients, SS-A – 2 patients, Rib.P-Prot – 1 patient. In DDLE and SCLE were identified autoantibodies RNP/Sm – 3 patients.
Conclusions. For a complete identification of autoantibodies it is necessary to use at least two diagnostic methods, for example, IIF and Western blot. Western blot method is used to verify the results of IIF method using Hep-2010 cells.
